IS THIS SUPPLEMENT REALLY THE FOUNTAIN OF YOUTH?
I am, without question, a “lifestyle doctor.” My passion and understanding is what is normal physiology, and how do we get there using diet, exercise and targeted supplementation. The holy grail of longevity is protecting the mitochondria (regardless of whether or not you remember these little organelles from high school biology…). One of the most powerful ways to do this is via activation of a family of genes (the SIRT family of genes) through a low calorie, high phytonutrient diet. There is, however, a phytonutrient that also activates the SIRT family of genes. It is resveratrol, the compound found in red wine, red grapes and peanuts. Not that this is designed to REPLACE healthy lifestyles, but can be used as an adjunct to these changes.

Coral calcium, Tums, dairy. The list of miracle compounds to increase bone density can be found easily on the Internet.
And if you read it on the Internet, it’s got to be true, right?
Just how many times has it been drilled into your brain that 3 servings of milk, cheese and yogurt per day will make your bones, teeth, nails and nasal hair stronger? Ok, so maybe you missed the nasal hair ad, but you get the point.
So, given all the hype (and BTW, yes the whole dairy = strong bones thing IS way more hype than reality), how can you piece together what’s good for bone health.
Luckily for you, on I’m a plane heading to dreary Chicago as I write this to teach a 12 hour class on the relationships between the gut, bone health and diabetes. So I couldn’t be more brushed-up on this topic.
Let’s get one thing out of the way first. Healthy bones are not about talking calcium supplements. This couldn’t be further from the whole story. Rather, you need to view this in the context of “bone health,” understanding that bone is not an inert tissue that we happen to attach our bones, tendons and ligaments to. Rather, bone tissue is an incredibly active tissue being affected by the health of your GI tract and, in turn, affecting your risk of diabetes. It also plays a critical role in producing immune cells, platelets and red blood cells. Do you really think calcium alone can handle this enormous task?
The list of things playing a positive or negative role in bone health are quite long, but the short list includes:
- Optimal levels of vitamin D
- Eating a plant-based diet loaded with natural calcium sources like green leafies
- Weight bearing exercise
- Avoiding drugs that affect serotonin levels (like SSRIs)
- Avoiding drugs that block acid production in the stomach (like Nexium)
The long list is enough to take up an entire eBook (which is in the development stages).
So what does all this have to do with this particular article? In it, researchers looked at the ability of a high dose of resveratrol (the protective compound found in red wine from the skin of the grapes as well as from peanut skins) to have an impact on bone density. They specifically looked at 74 middle-aged obese men who were pre-diabetic (Metabolic syndrome, or MetS) because being pre-diabetic is bad for your bones.
Here’s the details of the study:
- They were given either 1,000 mg or 150 mg of resveratrol a day or a placebo daily for 16 weeks.
- Bone alkaline phosphatase (BAP) was measured to look for how fast bone was being remodeled (higher = more bone turnover).
- BAP increased the most with the highest dose (around 4%).
- The volume of the bone in the lumbar spine increased 2.6% in the high dose group.
Before you go digging around in your “Principles of Bone Density” textbook you’ve right by your side, I can tell you can a 2.6% increase in bone volume in just 4 short months is pretty amazing. And all without breaking a single drop of sweat or drinking liquid from a cow’s udder meant to raise its young to an independent heifer.
Not quite in that same breath I can also tell you that 1,000 mg of resveratrol is a pretty high dose. It’s not that you’d have GI upset or grow a third limb; it’s just that this would be a pricey proposition to increase your bone density. This much resveratrol, from a good quality source, is likely going to run around $100 / month or more.
Of course, when compared to the cost of drugs used to treat bone density, it’s still a very cheap bet. As way of comparison, the newest drug used to treat osteoporosis, Denosumab (Xgeva), runs about $2,000 per monthly injection. Makes the resveratrol look like quite a bargain.

The idea of wine being healthy has been debated heavily. First, a clarification–red wine is the wine usually studied. Second, I firmly believe it is not the alcohol content, but rather, the bioflavanoids such as resveratrol from the red grapes (also found in peanuts) that offer the protective benefits. Considering that alcohol does increase your risk of certain types of cancer, skip the wine, eat lots of grapes and you’ll also get many other healthy substances and antioxidants.
Ann Intern Med. 2000;133:411-419 Wine may have protective effects beyond those associated with light alcohol consumption, according to findings of a pooled cohort study involving more than 24,000 subjects. The study is published in a recent issue of the Annals of Internal Medicine. The investigation included 13,064 men and 11,459 women, 20 to 98 years of age, who were participants in the Copenhagen City Heart Study, the Copenhagen Male Study, and studies from the Copenhagen County Centre of Preventive Medicine. Subjects reported their average weekly intake of wine, beer, and spirits. The researchers also took into account participants’ smoking status, education level, physical activity level, and body mass index. Compared with nondrinkers (whose relative risk was set at 1.0), light drinkers (those who consumed one to seven alcoholic drinks per week) had a relative risk of death from all causes of 0.82, but those who drank more than 35 drinks per week had a relative risk of 1.1. In general, alcohol consumption was associated with a reduced risk of death from coronary heart disease but an increased risk of death from cancer (especially in subjects who drank more than 21 drinks per week).Wine consumption at all levels — light, moderate, and heavy — was associated with a lower mortality rate in all three categories (all causes, cancer, and heart disease). Subjects who drank eight to 21 glasses of wine per week had a relative all-cause mortality risk of 0.76; from cancer, 0.78, and from heart disease, 0.64. Furthermore, compared with light drinkers who did not drink wine, light drinkers for whom wine made up 30% or more of their total alcohol intake had a 0.67 relative risk of all-cause mortality.Study authors Grønbæk et al speculate that “Wine may contain one or several substances that add to the beneficial effect of intake of a small amount of ethanol.”

This study on colon cancer cells shows an additive effect of resveratrol (found in red wine and peanuts) and butyrate (a short chain fatty acid formed by bacteria acting on soluble fiber in the gut). With studies like this, we start to see why certain traditional diets have the health -promoting effects they have. The Mediterranean diet has already show to be beneficial via increased absorption of lycopene (fat soluble carotenoid) with olive oil intake. This research study mechanism would find lower rates of colon cancer in patients combining wine intake with whole grains.

Resveratrol is a bioflavanoid found in peanuts and grapes that is believed to be the reason why wine is good for the heart.
This study suggests that we can add prostate cancer protection to the cardioprotective list.

My friends think I’m nuts, but I really am starting to believe that oxidative stress and subsequent mitochondrial dysfunction is darn close to the root of all evil. Here we see the link to enlargement of the heart and how this event is protected against by a synthetic version of resveratrol. Resveratrol is the compounds found in red wine, grapes and peanuts that is known to protect the heart.
My question is, we already know that resveratrol protects the heart–why are we looking for a synthetic version? Can you say patent…?
MECHANISM UNDERLYING OXIDATIVE STRESS – 05-01-05)
Mechanism underlying oxidative stress-mediated lipotoxicity
In a nutshell, the researchers in this study found that elevated triglycerides (as would be found in insulin resistance) get sucked up into the macrophages (a type of white blood cells), and once there, started to destroy the mitochondria of the cell resulting in cell death. The death of a macrophage in the bloodstream can lead to the production of foam cells and subsequent plague in the arteries.
Interestingly, vit C, NAC and resveratrol protected the macrophages from this death. So, to extrapolate into a living, breathing human, antioxidants protect the cells that produce plaquing in the arteries, resulting in less plaque.
